Brilliant Cresyl Blue Staining Enhances the Morphological Quality Evaluation of Porcine Oocytes-a Preliminary Study

چکیده

Abstract Brilliant cresyl blue (BCB) staining is a method used for selection of developmentally competent porcine oocytes, in addition to the standard morphological classification. The aim current study was assess relationship between BCB oocytes with high and low characteristics its maturation rate. In first part study, cumulus-oocytes complexes (COCs) (n=434) were aspirated from 60 ovaries. According classification staining, they divided 4 groups: COC+/BCB+, COC−/BCB+, COC+/BCB−, COC−/BCB−. second experiment, COCs (n=445) categorized into 3 control, BCB+, BCB−. Significant differences rate observed COC+/BCB+ COC−/BCB− (66% vs. 23%), COC−/BCB+ 38%) (p<0.0001). also registered COC−/BCB-(38% 23%) COC+/BCB− (53% (p<0.01). Results experiment showed that only BCB− had significantly lower (28%) compared control (63%) BCB+ (59%) (p<0.001). These findings higher developmental ability morphology. BCB-staining high-quality did not have significant impact on their group, but it might be useful selecting morphology characteristics.

Selection of Ovine Oocytes by Brilliant Cresyl Blue Staining

Sheep oocytes derived from the ovaries collected from the slaughterhouse are often used for research on in vitro embryo production, animal cloning, transgenesis, embryonic stem cells, and other embryo biotechnology aspects. Improving the in vitro culture efficiency of oocytes can provide more materials for similar studies. Generally, determination of oocyte quality is mostly based on the layers...

Brilliant Cresyl Blue in Sap of Nitella

It has been shown by several investigators I that when the pl-I value of the external dye solutions is increased, the rate of accumulation of a basic dye in the cell sap is accelerated. Two different explanations for this have been given. Some have accounted for it on the basis that the basic dye enters the cell in the form of a dye hydrate and combines with the substances in the cell; and that...

On-spot brilliant cresyl blue staining in fine needle aspiration, a novel technique for cytological diagnosis.

OBJECTIVE Application of on-spot staining with brilliant cresyl blue, a supravital stain, was investigated in fine needle aspirates from breast, lymph node and thyroid. STUDY DESIGN Patients with palpable breast lumps, lymph nodes and thyroid nodules were included in the study. Brilliant cresyl blue was used to stain the smears, which were evaluated for smear adequacy as well as cytological d...

Developmental competence of prepubertal goat oocytes selected with brilliant cresyl blue and matured with cysteamine supplementation.

The aim of this study was to assess the effect of oocyte selection using the brilliant cresyl blue (BCB) test plus the addition of cysteamine to the in vitro maturation (IVM) medium to improve the in vitro embryo development of prepubertal goat oocytes. The oocytes were exposed to 26 microM BCB and classified according to their cytoplasm coloration: BCB+ (oocytes with blue cytoplasm) and BCB- (...

P-100: Improved Efficiency of Somatic Cell Nuclear Transfer in Sheep by Persecution of Recipient Oocytes with Brilliant Cresyl Blue Staining

Background: The most important factor affecting the efficiency of somatic cell nuclear transfer (SCNT) is the initial quality of the oocyte. Since the abattoir-derived oocytes are heterogeneous in quality, selection of cohort fully grown/competent oocytes is necessary for proper development of SCNT embryos. Materials and Methods: Glucose-6-phosphate dehydrogenase (G6PDH) is actively expressed i...
